seqWell Launches Loaded-transposase Portfolio to Support Expanding Use of NGS-based Gene Editing Analysis Methods

Tagify™ off-the-shelf and custom loaded-transposase reagents provide reliability and scalability to tagmentation-based on- & off-target gene editing analysis

In addition, seqWell will offer loading of transposases with customer-specified oligonucleotides, including loading of their high-performance TnX transposase.

The Tagify portfolio provides a commercial source of fully quality-controlled, loaded transposases with a defined level of enzyme activity and payload verification. This eliminates the need of laboratories to purify, load, and characterize the final loaded transposase prior to use in their assays, freeing up laboratory resources and reducing a source of assay-to-assay variability.

“Assays to define the genome-wide on- and off-target activity of genome editors are important for both biomedical research and therapeutics,” said Shengdar Tsai, Ph.D, Associate Member, Department of Hematology, St. Jude Children’s Research Hospital. “Broadly adopted, sensitive, unbiased, biochemical and cellular methods of understanding the genome-wide activity of editors, such as CHANGE-seq and GUIDE-seq-2, rely on transposase-based tools. The commercial availability of standardized transposase reagents will help lower barriers for more labs to implement these assays rapidly and reliably.”

Tagify i5 UMI adapter-loaded transposase reagents support gene editing analysis assays, such as the UdiTaS™ method, that rely on tagmentation to simultaneously fragment and tag genomic DNA with Illumina-compatible adapters using a highly scalable workflow. These off-the-shelf reagents are available as a set of either 24 or 96 transposases each containing an i5 barcode and a unique molecular indentifier (UMI) for high throughput applications.

The Tagify custom-loading option enables scientists to address the growing number of NGS-based on- and off-target gene editing methods that rely on tagmentation via the flexibility to load either hyperactive Tn5 or seqWell’s recently introduced high performance TnX transposase, with Illumina-compatible barcodes, UMIs, universal adaptors, or a custom oligonucleotide sequence.
